Role for ¥âPIX in Migration of HCT116 Colon Cancer Cells

Abstract
Purpose: ¥âPIX is a regulator of cell migration and adhesion as well as a guanine nucleotide exchange factor. Therefore, ¥âPIX acts upstream of Rho GTPases and affects cell migration through reorganization of actin cytoskeleton. ¥âPIX has been reported to overexpress in breast cancer. In the present study, we sought to explore that ¥âPIX is overexpressed and regulates migration of HCT116 colon cancer cells. Materials and Methods: HCT116 cells were cultured to confluence and wound was applied. Migration was monitored until 22 hr following wound scratch. Analysis was performed using IncuCyte software. siRNA effect was analyzed by Western blot. Results: Expression levels of ¥âPIX in HCT116 cells were markedly down-regulated by ¥âPIX-specific siRNA but not control siRNA. PAK that makes a complex with ¥âPIX was not down-regulated by ¥â PIX-specific siRNA. Would was fully covered with migratory cells 22 hr following wound scratch. Contrary to our expectation, ¥âPIX siRNA-treated cell showed a faster rate in migration than control cells until 22 hr. Conclusion: It is suggested that ¥âPIX expression might regulate migration of HCT116 cells.
